Descriptif : The ubiquitin E3 ligase UBE3A is a critical enzyme required for appropriate brain development and function. In fact, Angelman syndrome (AS) and dup15q-associated autism are caused by the lack and excess of UBE3A in the brain, respectively. We recently found that UBE3A is responsible for ubiquitinating DDI1, among other proteins, in the brain but still there is a crucial gap in our understanding of the biological role of UBE3A in neurons. Aiming to fill that gap, in the present project we will focus on studying the type of UBE3A-dependent ubiquitination that occurs on DDI1 and revealing the biological effect of such modification in neurons. Characterising in vivo the substrates through which UBE3A neuronal activity is modulated will allow us to have a better understanding of the causes and symptoms related to Angelman and Dup15q syndromes, which confers to this ambitious project a multidisciplinary and transversal perspective. This is highly significant since at present there is no cure and no targetted treatment for either of those debilitating neurological disorders.
